Ligninolytic enzymes production and Remazol brilliant blue R decolorization by tropical brazilian basidiomycetes fungi BJM
Machado,Kátia M. G.; Matheus,Dácio R.; Bononi,Vera L. R..
Remazol Brilliant Blue R (RBBR) dye was used as substrate to evaluate ligninolytic activity in 125 basidiomycetous fungi isolated from tropical ecosystems. The extracellular RBBR decolorizing activity produced when selected fungi were grown in solid media and in soil contaminated with organochlorines was also evaluated. A total of 106 fungi decolorized the RBBR during the growth in malt extract agar (MEA, 2%); 96 fungi showed a mycelia growth and decolorization activity stronger than the P. chrysosporium used as reference. Extracellular extracts of 35 selected fungi grown on solid medium with sugar cane bagasse (BGS) were evaluated for RBBR decolorization and peroxidase activity. All fungi showed peroxidase activities, but 5 of those were unable to...

Screening of inducers for laccase production by Lentinula edodes in liquid medium BJM
Cavallazzi,José Renato P.; Kasuya,Catarina M.; Soares,Marcos A..
Laccases are enzymes involved in lignin degradation and are produced by various organisms. Due to their low substrate specificity their potential to be used in biotechnological applications has received attention. The addition of laccase inducers to the culture medium of microorganisms can enhance laccase production and facilitate its purification and utilization. The aim of this study was to investigate the effect of some compounds as laccase inducers in cultures of Lentinula edodes (shiitake). First, it was selected a culture medium suitable for laccase production by shiitake using two levels of N (2.6 and 26 mM) and seven levels of Cu (0, 50, 100, 150, 200, 250 and 300 µM). The medium with 2.6 mM N and 250 µM Cu was found to provide the highest laccase...

Diez J,M. Cristina; Gallardo A,Felipe; Saavedra,Gisselle; Cea L,Mara; Gianfreda,Liliana; Alvear Z,Marisol.
The activities of laccase, Mn-peroxidase, phosphatase and dehydrogenase were studied in a Chilean Andisol (Temuco Serie) contaminated with pentachlorophenol (PCP). Experiments were conducted on the top of 0-20 cm soil, adding different concentrations of PCP (0.41, 4.1 and 41 mg kg-1). Enzymatic activities and residual concentrations of PCP were evaluated in soil incubated at 25 ± 1 ºC for 112 days. Enzyme activities of PCP-treated soil varied according to the PCP concentration added. For instance, an inhibitory effect on Mn-peroxidase activity was not observed during incubation at the three levels of PCP, whereas acid phosphatase activity was only affected by the 0.41 mg kg-1 PCP treatment. By contrast, laccase and dehydrogenase activities were negatively...

Biodegradation of remazol brilliant blue R by ligninolytic enzymatic complex produced by Pleurotus ostreatus BJM
Machado,Kátia Maria Gomes; Matheus,Dácio Roberto.
Pleurotus ostreatus ("shimeji") is produced in Brazil on a commercial scale using various lignocellulosic residues. Efforts have been made to reuse the culture residue to obtain products of greater aggregate value such as enzymes or in processes of bioremediation. We evaluated the Remazol brilliant blue R (RBBR) degradation potential of extracts from solid substrate colonized by P. ostreatus and extracts from residue of the "shimeji" mushroom yield. Colonized substrates and residue were provided by Toyobo do Brasil Ltda. Extraction was performed with sodium acetate buffer (50 mM, pH 4.6). RBBR decolorization was monitored at 592 nm and peroxidase and laccase activities were measured by monitoring the oxidation of ABTS. Horseradish peroxidase was used as...

Trametes versicolor growth and laccase induction with by-products of pulp and paper industry Electron. J. Biotechnol.
Rebelo Barreto Xavier,Ana Maria; Mora Tavares,Ana Paula; Ferreira,Rita; Amado,Francisco.
The cultivation of Trametes versicolor for laccase production and cell growth were strongly dependent on experimental conditions namely physical and chemical parameters as well as nutrient availability and inducer stimulation. Biomass growth was compared for a rich medium and for a defined medium in two different temperatures. The best temperature was 28ºC and the maximum specific growth rates were µmax = 0.083 h-1 for the rich medium and µmax = 0.043 h-1 for the defined medium. It was clearly shown that laccase production is not associated with cell growth, indicating that this ligninolytic enzyme must be produced in the defined medium by a secondary metabolism. In order to obtain laccase induction, addition of solid lignin, lignosulphonates, veratryl...

Screening of Colletotrichum (Ascomycota) isolates, causal agents of Soybean Anthracnose, for Laccase production Boletín de la Sociedad
Levin,L.; Ramos,A .M.; Parisi,M .; Gally,M..
Colletotrichum truncatum is the most common pathogen fungus associated with soybean anthracnose. Although the lignin-degrading enzyme laccase has been implicated in pathogenicity of a wide range of plant pathogenic fungi, its biological role in the Colletotrichum -soybean disease system is unknown. The extent of the infection in our country led us to examine laccase production in Argentinean Colletotrichum strains isolated from diseased soybean plants from different geographic locations. Ten strains (eight of them identified as C. truncatum) , were screened for in vitro laccase production. Only six of the isolates, all of them C. truncatum , produced laccase activity when cultured on a defined medium based on pectin and asparagine as carbon and nitrogen...

Use of Laccase-Producing Microorganisms in Membrane Systems for Polluting Agents Removal: Consideration and Perspectives Chilean J. Agric. Res.
Serna C,Liliana; Cuesta A,Fernando L.
Contamination is defined as the presence of pollution agents in an ecosystem reaching an abnormal concentration level and causing adverse effects. Due to the important environmental problems that these cause in the food chain, diverse methods have been formulated to minimize those effects; among these enzymatic methods associated to microorganisms or other biological agents are used, and systems that allow to metabolize pollutants in an effective and practical way. The combined use of these systems appears to be the most appropriated option to eliminate pollution agents at a reasonable energy cost which allows their use in the industry. The present review identifies the adequate process conditions for technological implementation of biofilm-type membrane...

Pentachlorophenol removal by Pleurotus pulmonarius in submerged cultures BABT
Souza,Daniela Farani de; Costa,Silvio Cláudio da; Dacome,Antonio Sérgio; Souza,Cristina Giatti Marques de; Bracht,Adelar; Peralta,Rosane Marina.
Pentachlorophenol (PCP) removal by Pleurotus pulmonarius grown in submerged cultures in the presence and absence of laccase inducers was studied in this work. When PCP was added to a final concentration of 25 mg·L-1 in submerged cultures actively producing laccase, 70% of the PCP was removed after 96 h. The removal of PCP was less than 20% in the cultures with low laccase activity. The results suggested that laccase played an important role in the biodegradation of PCP by P. pulmonarius and that for bioremediation purposes the fungus must be cultured under the conditions of active laccase production.
